This project aims to explore a fluorogenic reaction, namely the synthesis of indolizines via a 1,3-dipolar cycloaddition between a pyridinium and an alkyne, as a transduction mechanism for the recognition between an aptamer and its target. Several indolizine derivatives have been synthesized, characterized, and used to label oligonucleotides. Fluorescence and duplex stability studies have demonstrated the potential of indolizines as novel DNA biomarkers. Unfortunately, when both partners (the alkyne and the pyridinium) are attached to two complementary DNA strands, the reaction does not proceed because a constituent element of DNA interferes with it; its identification is ongoing. Other indolizine-based fluorogenic reactions are currently being investigated.
